Supplementary Fig 1. Schematic of the glymphatics-on-chip platform and hydrostatic setup. (a) Top-down view (X-Y plane) of the microfluidic chip layout, consisting of two 6 mm-diameter media reservoirs (red and blue) connected by a central hydrogel chamber and flanked by two hydrogel casting ports. The full chip footprint measures 2.7 cm × 2.0 cm. (b) Cross-sectional side view (X-Z plane) illustrating the PDMS enclosure bonded to a glass coverslip. Two 250 µm-diameter microchannels border the hydrogel region and enable perfusion across the cell-laden ECM. (c) Cross-section showing the hydrostatic pressure configuration used in drainage assays. Reservoirs were filled with 200 µL and 40 µL of media + dextran, respectively, creating a pressure gradient of ~62.5 Pa. (d) Cross-section side view of the ECM reservoir, showing a gel height of 250 µm and the direction of interstitial fluid flow from left to right. (e) Top-down view (X-Y plane) of the ECM reservoir, showing a gel width of 3 mm and unidirectional fluid flow across the ECM at ~1.7 µm/s.
